I often struggle to take off at just the right speed. Does anyone know how to calculate your rotation speed?

Usually 150 kts is good.

You could also trial and error it yourself because there are almost an infinite amount of variations affecting your Vr.
First find out your Vmu (minimum unstick speed) for whatever weight you are and what thrust setting you want to use by performing a take off with full back stick and note what airspeed your main wheels come off the ground. Vr must be at least 110% of that speed.
Test flying 101.
